雌二醇ELISA試劑盒是一種固相酶聯免疫吸附試驗（ELISA），基于競爭性結合的原理。微孔板孔被涂覆有針對雌二醇分子抗原位點的多克隆[兔]抗體。

在第一次孵育過程中，添加的樣本中的雌二醇與添加的酶聯物競爭結合，后者是與辣根過氧化物酶結合的雌二醇，以結合到涂覆的抗體上。經過洗滌步驟去除所有未結合的物質后，將固相與底物溶液孵育。通過添加終止溶液來停止比色反應，測量所得黃色產物的光密度（OD）。顏色的強度與樣本中分析物的濃度成反比。

通過繪制標準品濃度對OD值的標準曲線，使用該標準曲線確定未知樣本的濃度。
