犬尿氨酸是一種非蛋白質性氨基酸,在色氨酸降解過程中作為代謝中間體產生[1-5]。色氨酸的降解是由誘導型酶吲哚胺-2,3-雙加氧酶(IDO)催化的。產品為犬尿氨酸[4,6-8]。細胞因子,特別是干擾素γ[5,9,10],會影響IDO的活性,因此犬尿氨酸途徑與免疫系統密切相關[9,11]。犬尿氨酸可以進一步轉化為具有神經保護作用的犬尿氨酸酯,也可以轉化為具有神經毒性的喹啉酸[6,11]。

LDN Labor Diagnostika Nord-犬尿氨酸ELISA預期用途和原理:
酶免疫測定法用于定量測定血清和EDTA血漿樣品中的L-犬尿蛋白,以評估L-犬尿素穩態。在酰化過程中,犬尿蛋白在37°C下被激活,隨后與蛋白質偶聯。隨后的競爭ELISA使用微量滴定板形式。抗原與微量滴定板的固相結合。酰化標準品、對照品和樣品的分析物濃度與固定數量的抗體結合位點的固相結合分析物濃度相匹配。在系統處于平衡狀態后,通過洗滌除去游離抗原和游離抗原-抗體復合物。通過偶聯TMB作為底物的抗兔IgG過氧化物酶檢測與固相結合的抗體,從而產生顯色反應。在450nm的波長下監測反應。通過將未知樣品的吸收與用已知標準濃度制備的參考曲線進行比較來實現未知樣品的定量。建議手動處理ELISA。自動實驗室設備的使用由用戶負責。本產品不用于臨床診斷。
