超臨界流體色譜技術 (Supercritical fluid chromatography, SFC) ,顧名思義,即流動相既不是氣體也不是液體,而是使用超過臨界溫度和臨界壓力下的單一態流體(Supercritical Fluid)作流動相進行分離分析、制備純化的色譜技術。
SFC的原理與液相色譜的原理類似,然而利用超臨界流體(通常為二氧化碳CO2)取代有機溶劑或水作為主要流動相,從本質上來說是一種高效的正相色譜技術。作為流動相所使用的超臨界流體CO2具有獨特的低粘度和高擴散性能而成為一種智能溶劑,因具有與液體相近的密度,對各種物質有較強的溶解性,難揮發性和熱穩定性差的物質分析均適用;同時,超臨界流體的粘度近于氣體,比液體約低1~2個數量級,而它的擴散系數在氣體和液體之間,因而具有較快的傳質速度和較小的傳質阻力,分離過程中峰形變窄、柱效增加、分析速度加快。由此可見,SFC揚HPLC與GC之長、避GC及HPLC之短,具有獨特的優越性,應用前景極為廣闊。
由于SFC采用天然“綠色”的超臨界流體CO2作為溶劑,無需有機溶劑回收處理,省事經濟環保,節能降耗又可持續使用,因而SFC對生物醫藥、精細化工、生態環境、農業食品等行業具有獨特的吸引力。
SFC在制藥行業,尤其是手性藥物開發領域,由于其能夠有效地利用液相色譜中常用的手性固定相 (chiral stationary phase,CSP) 且與GC、HPLC體系中使用的檢測器完全兼容, SFC是一種理想的手性分離方法。因為它運行速度更快,溶劑用量更少,樣品后處理更簡單,總體上來講,SFC是一種更經濟、更綠色的方法,在手性化合物分離方面性能卓越,大有超越高效液相色譜法(HPLC)之趨勢,為分離純化和手性制備分離的有益替代方案。
