Bispecific antibodies can be applied for a broad range of diagnostic and therpaeutic applications, e.g., the retargeting of effector molecules and cells to tumor cells or other structures. This protocol describes the generation of bispecific diabodies and single-chain derivatives thereof from the variable heavy and light chain domains of two antibodies and provides some information for the analysis of bispecific binding.
